After Graeme Smith and Alviro Petersen successfully negotiated the new ball, Ajmal proved the spinners could have an important role to play in a match still only in its second day.
The 35-year-old successfully used DRS to send Smith back to the pavilion after missing a sweep before Petersen was well caught by Azhar Ali at short leg. Hashim Amla (17) and Faf du Plessis (three) remained at tea as South Africa nervously progressed to 65 for two.
