Right-back De Laet (ankle) and midfielder James (knee) were absent prior to the international break but the two-week sabbatical has seen them recover.
Winger Anthony Knockaert is also available again after a family bereavement.
Chris Wood's early return from international duty with New Zealand is an added bonus for Pearson and the striker is likely to start against the club where he scored 11 goals in 19 appearances during a loan spell earlier this season.
Wood had been a major doubt for the game on Friday night. He was due to play for the All Whites in their World Cup qualifier in the Solomon Islands on Tuesday and would have struggled to be ready to face Millwall after a 36-hour return flight.
However, New Zealand secured their place in the play-offs for Brazil 2014 with victory against New Caledonia last weekend and Wood was released to return ahead of schedule, arriving back at Leicester on Monday.
Goalkeeper Kasper Schmeichel (Denmark) defender Sean St Ledger (Republic of Ireland) and midfielder Andy King (Wales) have also returned unscathed from international duty and will be available.
