With Stuart Beavon (knee) the only player unavailable, Grayson has a strong squad at his disposal but players such as Will Hayhurst and Iain Hume, who came on for the closing stages of Saturday's goalless draw against Wolves, may be in line to start.
"Any success we are going to have this season is going to be as a squad and not as individuals," Grayson told the club's official website ahead of the clash.
Grayson spent six years at Blackpool both as a player and manager.
The clash has been labelled 'All White On The Night', with fans urged to create a sea of white in the stands and with it an intimidating atmosphere.
Blackpool boss Paul Ince has a number of options should he wish to make changes.
Three of the Seasiders' summer signings started Saturday's Sky Bet Championship opener at Doncaster, with defender Gary MacKenzie scoring the second goal in a 3-1 win.
Forwards Steven Davies and Bobby Grant were also included from the start, while Michael Chopra, Jack Robinson and Neal Bishop were on the bench. They may feature more prominently at Deepdale.
Chris Basham may also have played his way into the starting line-up after earning the praise of his manager for an impressive substitute display at the Keepmoat.
