The former Chelsea manager denied any contact with City in his pre-match press conference ahead of Saturday's trip to Osasuna in La Liga.
Asked about stories in the Spanish press linking him with the Etihad Stadium, Mourinho said simply: "I can reject (them)."
City's Premier League title bid has stalled in recent weeks, as they have lost a five-point lead over Manchester United to slip three points behind their neighbours.
Although Real are six points clear at the top of La Liga, their poor record in matches against Barcelona, and a perception of arrogance on Mourinho's part, have meant the Portuguese is expected to leave at the end of the season.
Mourinho won two Premier League titles and an FA Cup with Chelsea before his sacking in 2007.
